Why are salt and alcohol used together in DNA extraction?

Answer and explanation

Correct answer: To precipitate and separate DNA

The phosphate groups of DNA are negatively charged. Cations from salt partly neutralise this charge, while alcohol lowers DNA solubility. Consequently, DNA forms a precipitate that can be separated. Alcohol alone may precipitate DNA less effectively because of the remaining charge repulsion. Exam tip: In DNA extraction, salt plus cold alcohol is mainly used for DNA precipitation.
