What problem can occur by reusing buffer in restriction digestion gel?

Answer and explanation

Correct answer: Poor conductivity and weak band resolution

In gel electrophoresis, repeatedly used running buffer can become depleted of ions and its pH may change. This lowers conductivity and can produce faint or poorly resolved DNA bands. Reusing buffer does not increase the amount of restriction enzyme. Exam tip: use fresh buffer of the correct concentration for clear bands and proper migration.
