Why should a restriction enzyme not be vigorously vortexed?

Answer and explanation

Correct answer: There is a risk of damage to the enzyme protein or loss of its activity

Restriction enzymes are proteins. Vigorous vortexing can create shear forces and foaming that may disturb the enzyme’s structure and reduce its ability to cut DNA. The buffer does not evaporate merely because of vortexing; enzymes are usually mixed gently by pipetting or by gently flicking the tube. Exam tip: In enzyme handling, “gentle mixing” helps preserve enzyme activity.
