What problem occurs if a small fragment runs out of the gel in restriction digest?

Answer and explanation

Correct answer: The desired DNA band may be lost

Small DNA fragments migrate faster and farther through a gel. If electrophoresis is continued for too long, a small fragment can run off the lower end of the gel, so its desired DNA band will no longer be visible or recoverable. This does not increase the amount of restriction enzyme. Exam tip: a higher-percentage agarose gel and a shorter run time help retain small fragments.
