Why is DNA concentration measured again after restriction-digestion cleanup?

Answer and explanation

Correct answer: DNA may be lost during cleanup

The governing concept is quantitative recovery after purification. A cleanup step removes restriction enzymes, salts, buffer components, and small molecules, but recovery is not necessarily complete. Some DNA may remain on the column, be lost during transfers, or be eluted in a smaller or larger volume than expected. Measuring concentration again gives the actual amount available for the next reaction. This value is important for calculating the DNA volume and insert-to-vector ratio used in ligation; relying on the starting concentration could lead to too little or too much DNA. The measurement is not intended to show that DNA doubles, to replace DNA quantification with enzyme concentration, or to determine gel colour.
