Why can fragment size estimation be wrong in restriction digestion?

Answer and explanation

Correct answer: Due to an unsuitable agarose gel percentage or an incorrect DNA ladder

DNA fragment size is estimated in agarose gel electrophoresis by comparing its migration with bands of known size in a DNA ladder. An unsuitable gel percentage or a ladder with an inappropriate size range can give poor separation and an incorrect size estimate. The colour of a restriction enzyme and the taste of water do not affect DNA migration. Exam tip: use a higher agarose percentage for smaller fragments and a lower percentage for larger fragments.
