A vector has a selectable marker but no ori. What problem will occur in the host?

Answer and explanation

Correct answer: The vector will not replicate autonomously in the host cell

The ori (origin of replication) is the DNA sequence from which vector replication begins. Without an ori, the vector cannot replicate autonomously in the host cell and will not be stably maintained as cells divide. A selectable marker may help in selection, but it cannot compensate for the absence of ori. Exam tip: remember that ori is required for replication, whereas a selectable marker is used to identify or select transformed cells.
